Создан новый фототоксичный белок

Ученые из Сколтеха, Института биоорганической химии РАН и Института медицинских наук в Лондоне разработали улучшенную версию фототоксичного белка SuperNova, генетически кодируемого белка, позволяющего влиять на внутриклеточные процессы при воздействии света. Исследователи ожидают, что белок можно будет применять к широкому кругу экспериментальных моделей в качестве генетически кодируемого фотосенсибилизатора. Разработка важна для медицинских целей.

Результаты исследования опубликованы в журнале International Journal of Molecular Sciences. Фототоксичные белки – важный инструмент для ученых. Такие белки используют как генетически кодируемые фотосенсибилизаторы, чтобы генерировать активные формы кислорода при облучении светом.

В отличие от обычных химических фотосенсибилизаторов, фототоксичные белки кодируются генетически и экспрессируются самой клеткой, что позволяет легко ими управлять и направлять в любой выбранный участок клетки.

За счет образующихся под действием света активных форм кислорода фототоксичные белки могут создавать строго локализованный окислительный стресс, например, для уничтожения выбранной клеточной популяции и инактивации целевых белков, что особенно нужно при создании моделей для изучения клеточных процессов.

Первый фототоксичный белок был описан российскими учеными под руководством профессора Центра наук о жизни Сколтеха Константина Лукьянова в 2006 году, его назвали KilllerRed, позже японские ученые усовершенствовали его и назвали SuperNova. В своей новой работе ученые под руководством Константина Лукьянова предлагают улучшенную версию белка SuperNova – SuperNova2.

Новый белок сочетает в себе высокую скорость и полноту созревания и мономерное состояние, что облегчает использование и делает его подходящим для широкого спектра молекулярно-биологических задач. «Мы ожидаем, что белок SuperNova2 будет применим к широкому кругу экспериментальных моделей в качестве генетически кодируемого фотосенсибилизатора», – комментирует профессор Константин Лукьянов.
